Inhibitory Effect of Arctigenin on the Virulence Factors and Growth of Helicobacter pylori

Objectives: Helicobacter pylori infection in the human stomach causes various gastrointestinal diseases, including gastritis, gastric ulcer, and gastric cancer. Arctigenin, a lignan extracted from the greater burdock Arctium lappa, has been reported to possess antiviral, antibacterial, anti-inflammatory, and anticancer activities. Methods: In this study, the antibacterial effect of arctigenin against H. pylori was confirmed using the agar and broth dilution methods. Additionally, changes in mRNA expression were assessed using reverse transcription-polymerase chain reaction to investigate the effect of arctigenin on the virulence factors of H. pylori. Results: The minimum inhibitory concentration of arctigenin against H. pylori was found to be 1,340 nM. Arctigenin inhibited H. pylori growth by downregulating holB, polA, and rpoN, which are crucial components of the bacterial replication and transcription machinery. Conclusion: Arctigenin also inhibited urease activity and the expression of flagellar components in H. pylori, suggesting that it may prevent H. pylori from successfully colonizing the gastric mucosa. Furthermore, cytotoxin-associated protein A and vacuolating cytotoxin A, the most important virulence factors in the pathogenesis of H. pylori, as well as their secretion systems, the type IV secretion system and type Va secretion system, were significantly inhibited by arctigenin. Taken together, these findings provide scientific evidence supporting the use of arctigenin for the prevention and treatment of H. pylori infection.
