Protecting Effects of Schisandra chinensis (Turcz.) Baill Seed Extract on PC12 Cells

Objectives: This study investigated the anti-inflammatory effect, related mechanism, and suppression of acetylcholinesterase (AChE) activity by Schisandra chinensis (Turcz.) Baill seed extracts (SCSE) treatment. In particular, we aimed to examine whether SCSE could modulate neuroinflammatory pathways relevant to cognitive decline. Methods: A soluble tetrazolium salt colorimetric assay was applied to SCSE-treated PC12 cells to examine cell viability. We measured inflammation-related factors and associated mechanisms via Griess reagent assay, Western blotting and reverse transcriptase-polymerase chain reaction in PC12 cells induced by lipopolysaccharide. Results: SCSE reduced activation of the toll-like receptor 4 signaling pathway-related proteins myeloid differentiation primary response 88, IκB kinase beta, and nuclear factor kappa B. It also reduced the activation of mitogen-activated protein kinase pathway-related proteins p38, extracellular signal-regulated kinase, and c-Jun N-terminal kinase and the inflammatory cytokines tumor necrosis factor-alpha, interleukin-1 beta, and interleukin-6. In addition, it suppressed nitric oxide production and reduced the related proteins cyclooxygenase-2 and inducible nitric oxide synthase. Finally, SCSE also reduced AChE activity by about 20%. Conclusion: These results suggest that the anti-inflammatory effect and suppression of AChE activity by SCSE prevent the progression of memory decline and help improve cognitive function, and further research is needed to utilize them as potential drugs.
