Effects of hyaluronic acid and activated platelet-rich plasma in canine chondrocytes

This study was performed to evaluate the proliferation, gene expression of extra-cellular matrix and anti-inflammatory effects of hyaluronic acid and platelet-rich plasma (PRP) in inflammation-induced canine chondrocytes. The inflammatory state was induced by 10 μg/mL of lipopolysaccharide for 6 hours. Four experimental groups were divided: control group, HA group, PRP group, and HA+PRP group. Each group was separately applied with no serum (control group), 100 μg/mL hyaluronic acid (HA group), 5% PRP (PRP group), or 100 μg/mL hyaluronic acid with 5% PRP (HA+PRP group) for 24 hours. The mRNA expressions of COL1A1, COL2A1, tumor necrosis factor (TNF)-alpha and aggrecan were compared through real-time chain polymerization (PCR). In the proliferation experiment, there were significant increases in PRP group and HA+PRP group. In real-time qPCR analysis, PRP group and HA+PRP group showed significant reduction on expression of COL1A1, COL2A1, aggrecan and TNF-alpha. PRP on canine chondrocytes had a positive effect on the proliferation and the inhibition of inflammation. When hyaluronic acid was applied simultaneously with platelet-rich plasma, it did not appear to inhibit chondrocyte proliferation and anti-inflammatory action in this study.
