The implications of supernumerary teeth to understand the developmental mechanisms underlying tooth formation

Dental anomalies are diverse. The most common clinical anomaly interfering with the tooth numbering system is supernumerary teeth (ST). Teeth are created by the reciprocal interactions of the surface epithelium ectoderm and the neural crest-derived mesenchyme regulated by numerous signaling molecules including SHH, WNT, BMP, FGF, and Eda. Any alteration or disruption to these signaling molecules may lead to severe tooth malformations. Various mice models have been used to investigate the related etiology and potential candidate genes involved in the pathogenesis of ST. ST can occur as sporadic conditions but are mostly associated with genetic syndromes. An understanding of the molecular basis of ST and the associated syndromes may help clinicians in early diagnosis and constructive treatment planning. The findings of this literature review on ST contribute to further understanding of tooth development and regeneration.
